- He played the role of Veerabhadran in the Tamil-language action film Captain Prabhakaran in 1991.
- He contested from Periyakulam as a candidate of the Puthiya Tamilagam (PT) in the 1999 Indian general election in Tamil Nadu and finished in the 3rd position after securing around one lakh votes.
- Mansoor Ali Khan was cast as McDowell Mani in the 2004 Telugu-language action film Samba, starring Jr. NTR, Bhumika Chawla, Genelia D’Souza, and Prakash Raj.
- He contested as an independent candidate at the 2009 Indian general election in Tamil Nadu. Criminal charges were pressed against him by the opposition for moving around in a vehicle with a propaganda banner breaching the moral conduct code.
- In January 2012, Mansoor Ali Khan was arrested on land grabbing charges after it was alleged that he had illegally constructed a 16-storeyed property in Arumbakkam.

- In an interview, Mansoor Ali Khan stated that he expected to do a rape scene with his Leo co-actor Trisha but didn’t even get a chance to share the screen with her. He received major criticism for his statement and Trisha replied that she found the statement “s*xist, disrespectful, misogynistic, repulsive and in bad taste”. He later apologized to Trisha and she responded by saying “To err is human, to forgive is divine”.
